The NADPH-dependent cytochrome P450 reductase (CPR) is a membrane bound protein localized in the ER membrane. CPR is a multidomain protein with three cofactor binding domains (FMN, FAD and NADPH) and a linker domain situated between the FMN and FAD/NADPH domains. CPR donates electrons from the two-electron donor NADPH to the heme of P450 in a coupled two-step reaction as illustrated in picture below. NADPH binds to the NADPH-binding domain and electrons are shuttled from NADPH through FAD and FMN to the P450 heme.
